Placental protein 13 (PP13, also known as galectin-13) is a protein that in humans is encoded by the LGALS13 gene.{{cite journal | vauthors = Bohn H, Kraus W, Winckler W | title = Purification and characterization of two new soluble placental tissue proteins (PP13 and PP17) | journal = Oncodevelopmental Biology and Medicine | volume = 4 | issue = 5 | pages = 343–50 | date = Jul 1983 | pmid = 6856484 }}{{cite journal | vauthors = Than NG, Sumegi B, Than GN, Berente Z, Bohn H | title = Isolation and sequence analysis of a cDNA encoding human placental tissue protein 13 (PP13), a new lysophospholipase, homologue of human eosinophil Charcot-Leyden Crystal protein | journal = Placenta | volume = 20 | issue = 8 | pages = 703–10 | date = November 1999 | pmid = 10527825 | doi = 10.1053/plac.1999.0436 }}

Structure and function

It is composed of two identical subunits which are held together by disulfide bonds. The monomer of this protein has structural similarity to several members of the beta-galactoside-binding S-type lectin family, but it could not bind beta-galactoside. This is because the ligand binding site is lack of key residue for binding beta-galactoside. {{cite journal | vauthors = Su J, Wang Y, Si Y, Gao J, Song C, Cui L, Wu R, Tai G, Zhou Y | title = Galectin-13, a different prototype galectin, does not bind β-galacto-sides and forms dimers via intermolecular disulfide bridges between Cys-136 and Cys-138 | journal = Scientific Reports | volume = 8 | issue = 1 | pages = 980 | date = January 2018 | pmid = 29343868 | doi = 10.1038/s41598-018-19465-0 | pmc = 5772480 | bibcode = 2018NatSR...8..980S }}It is a galectin-like protein.

Galectin-13 has been shown to bind CD44, inhibiting membrane localization of SLC7A11 and promoting propagation of ferroptosis.{{Cite journal |last=Zhang |first=Hai-Liang |last2=Guo |first2=Yi-Qing |last3=Liu |first3=Shan |last4=Ye |first4=Zhi-Peng |last5=Li |first5=Li-Chao |last6=Hu |first6=Bing-Xin |last7=Li |first7=Zhi-Ling |last8=Chen |first8=Yu-Hong |last9=Feng |first9=Gong-Kan |last10=Shen |first10=Hui-Qi |last11=Deng |first11=Rong |last12=Zhu |first12=Xiao-Feng |date=2025-04-17 |title=Galectin-13 reduces membrane localization of SLC7A11 for ferroptosis propagation |url=https://www.nature.com/articles/s41589-025-01888-2 |journal=Nature Chemical Biology |language=en |pages=1–13 |doi=10.1038/s41589-025-01888-2 |issn=1552-4469|url-access=subscription }}

Clinical significance

PP13 levels that are low in the first trimester of pregnancy confers a higher risk for developing pre-eclampsia later in pregnancy.{{cite journal | vauthors = Huppertz B, Meiri H, Gizurarson S, Osol G, Sammar M | title = Placental protein 13 (PP13): a new biological target shifting individualized risk assessment to personalized drug design combating pre-eclampsia | journal = Human Reproduction Update | volume = 19 | issue = 4 | pages = 391–405 | date = February 2013 | pmid = 23420029 | doi = 10.1093/humupd/dmt003 | doi-access = free }}
