Protein Serine/threonine-protein kinase H2 is a Ser/Thr protein pseudokinase that is encoded by the PSKH2 gene, and forms part of the CAMK subfamily of protein kinases. Homologues of PSKH2 are absent in rodents, but a PSKH2-like sequence is present in most vertebrate genomes.{{cite journal | vauthors = Byrne DP, Shrestha S, Daly LA, Marensi V, Ramakrishnan K, Eyers CE, Kannan N, and Eyers PA | title = Evolutionary and cellular analysis of the 'dark' pseudokinase PSKH2 | journal = Biochemical Journal | volume = 480 | issue = 2 | pages = 141–160 | date = Jan 2023 | pmid = 39964718 | pmc = 11873932 | doi = 10.1042/BCJ20220474 }} PSKH2 does not bind to ATP, nor does it phosphorylate any known substrate in cells, simplistically because it lacks the catalytic Asp residue in the HRD motif. The PSKH2 protein interactome is complex, and involves a number of membrane-associated proteins alongside the HSP90 Cdc37 molecular chaperone complex.{{cite journal | vauthors = Shrestha S, Byrne DP, Harris JA, Kannan N, and Eyers PA | title = Cataloguing the dead: breathing new life into pseudokinase research. | journal = FEBS J | volume = 287 | issue = 19 | pages = 4150–4169 | date = Oct 2020 | pmid = 32053275 | pmc = 7586955 | doi = 10.1111/febs.15246 }} PSKH2 exhibits significant sequence homology to the catalytically active Ser/Thr kinase PSKH1.{{cite journal | vauthors = Horne CR, Young SY, Mather LJ, Dagley LF, Johnson JL, Yaron-Barir TM, Huntsman EM, Daly LA, Byrne DP, Cadell AL, Ng BH, Yousef J, Multari DH, Shen L, McAloon LM, Manning G, Febbraio MA, Means AR, Cantley LC, Tanzer MC, Croucher DR, Eyers CE, Eyers PA, Scott JW, and Murphy JM | title = PSKH1 kinase activity is differentially modulated via allosteric binding of Ca2+ sensor proteins | journal = Proceedings of the National Academy of Sciences USA | volume = 122 | issue = 8 | date = Feb 2025 | pages = e2420961122 | pmid = 39964718 | pmc = 11873932 | doi = 10.1073/pnas.2420961122 }}
