He successfully ran for governor of Bukidnon for the 2022 election where the successor of outgoing governor Jose Maria Zubiri Jr. was to be determined. He ran under the Nacionalista Party. Roque's rival was governor Zubiri's son and third district representative Manuel Zubiri.

As governor, Roque vetoed the passage of an indigenous people's code being proposed in the provincial board in August 2024, which led to criticism by the Provincial Tribal Council and threats by the leaders of Bukidnon's seven major tribes to have him declared persona non grata in their domains.{{cite news |last1=Rosete |first1=Franck Dick |title=Bukidnon governor faces backlash from province’s 7 tribes after veto of IP code |url=https://www.rappler.com/philippines/mindanao/bukidnon-governor-rogelio-neil-roque-backlash-veto-ip-code/ |access-date=7 August 2024 |work=Rappler |date=7 August 2024}}
