Thiazole synthase ({{EC number|2.8.1.10}}, thiG (gene)) is an enzyme with systematic name 1-deoxy-D-xylulose 5-phosphate:thiol sulfurtransferase.{{cite journal | vauthors = Park JH, Dorrestein PC, Zhai H, Kinsland C, McLafferty FW, Begley TP | title = Biosynthesis of the thiazole moiety of thiamin pyrophosphate (vitamin B1) | journal = Biochemistry | volume = 42 | issue = 42 | pages = 12430–8 | date = October 2003 | pmid = 14567704 | doi = 10.1021/bi034902z }}{{cite journal | vauthors = Dorrestein PC, Zhai H, McLafferty FW, Begley TP | title = The biosynthesis of the thiazole phosphate moiety of thiamin: the sulfur transfer mediated by the sulfur carrier protein ThiS | journal = Chemistry & Biology | volume = 11 | issue = 10 | pages = 1373–81 | date = October 2004 | pmid = 15489164 | doi = 10.1016/j.chembiol.2004.08.009 | doi-access = free }}{{cite journal | vauthors = Dorrestein PC, Zhai H, Taylor SV, McLafferty FW, Begley TP | title = The biosynthesis of the thiazole phosphate moiety of thiamin (vitamin B1): the early steps catalyzed by thiazole synthase | journal = Journal of the American Chemical Society | volume = 126 | issue = 10 | pages = 3091–6 | date = March 2004 | pmid = 15012138 | doi = 10.1021/ja039616p }}{{cite journal | vauthors = Settembre EC, Dorrestein PC, Zhai H, Chatterjee A, McLafferty FW, Begley TP, Ealick SE | title = Thiamin biosynthesis in Bacillus subtilis: structure of the thiazole synthase/sulfur carrier protein complex | journal = Biochemistry | volume = 43 | issue = 37 | pages = 11647–57 | date = September 2004 | pmid = 15362849 | doi = 10.1021/bi0488911 }}{{cite journal | vauthors = Hazra A, Chatterjee A, Begley TP | title = Biosynthesis of the thiamin thiazole in Bacillus subtilis: identification of the product of the thiazole synthase-catalyzed reaction | journal = Journal of the American Chemical Society | volume = 131 | issue = 9 | pages = 3225–9 | date = March 2009 | pmid = 19216519 | pmc = 2765510 | doi = 10.1021/ja806752h }}{{cite journal | vauthors = Hazra AB, Han Y, Chatterjee A, Zhang Y, Lai RY, Ealick SE, Begley TP | title = A missing enzyme in thiamin thiazole biosynthesis: identification of TenI as a thiazole tautomerase | journal = Journal of the American Chemical Society | volume = 133 | issue = 24 | pages = 9311–9 | date = June 2011 | pmid = 21534620 | pmc = 3116082 | doi = 10.1021/ja1110514 }} This enzyme catalyses the following chemical reaction

: 1-deoxy-D-xylulose 5-phosphate + 2-iminoacetate + thiocarboxy-adenylate-[sulfur-carrier protein ThiS] \rightleftharpoons 2-[(2R,5Z)-2-carboxy-4-methylthiazol-5(2H)-ylidene]ethyl phosphate + [sulfur-carrier protein ThiS] + 2 H2O

H2S can provide the sulfur in vitro.
